depending on the age of the kids...

mine's 14 and recently we did the take a tram to acland street and have coffee and cake ( which you CAN really do anywhere BUT acland street has so much character) we then wandered around FED SQUARE and found a media museum we didn't know was there. We often explore the city and find interesting things,
Last Year we went on the Ghost Tour of melbourne after seeing a spooky play called *the woman in Black*

BUT if your kids are younger .. many of the things others have answered are great. I'd add healesville sanctuary and also a ride on Puffing Billy , Collingwood's Children's Farm .

AND if your up for an interesting adventure .. Try a* Head's or Tails* tour .. you start in the middle of the city and you flip a coin as to what direction tram you are going to catch .. you allow a flip of a coin to direct you and you could end up anywhere ,,discovering your own fun places in Melbourne

Luna Park in St Kilda, Geelong Adventure park, Melbourne Aquarium and Melbourne Zoo, Imax, Museum, Boat ride on the Yarra... how old are the kids?? Shopping @ chadstone is awesome... there heaps of indoor playgrounds around and if your interested in march Melbourne has this awesome festival called moomba, there are heaps of rides and entertainment for the whole family!
Lock them in the car and go to the Casino... just kidding but it has been done!

I'd take them to the Zoo. One of the best on the planet.
My niece came to visit at Easter and we had a ball!
Zoo, Aquarium, Museum, ACMI (fed squ), Cook's cottage, and we went down to see the seals and penguins at phillip island.
There's also heaps of shopping and just looking around. There's a free sightseeing tram you can take from flinders st station
Try the St Kilda precinct, take a tram ride there, visit Luna Park, take a stroll along Acland St , buy ice-creams, have a foot massage at Sole Sisters - fun for all the family ....and not an expensive day out.
