You also have to change trains at the border as Germany has a different rail gauge from Czech Republic. It doesn't look far, but Czech trains are very unreliable and usually late. I'd say hiring a car is easier and quicker. Taking the motorway border crossing signposted "Prag/Pilsen" from Munich and then turning north at A6/Junction 128 passing by Marienbad/Marianske Lasne is quicker than crossing by Cheb further north. I've also driven to Prague from the UK and France using these routes as well as living in Prague for a year and visiting Karlsbad/Karlovy Vary by car from there using the route I've recommended to you.
