It would probably be faster to take the greyhound bus from the bus station (bay/dundas) to U of G. The buses run more often, and it is a shorter ride- unless you get an express route from York Mills which I'm not sure exists. If you take the greyhound- it goes directly to Guelph, whereas, the GO Bus usually goes through a bunch of different places and can involve changing buses.

You could also take the VIA Train from Union Station. It is more expensive, and only runs twice a day. Once you get to to the train station- you would still have to get to campus- which would be about a $7-8 cab ride.
Others

You can take Rapid Transit from Finch station down to York Mills station. From there, take the Go Bus to Guelph.
