taking the sea bus over to Lonsdale Quay is good too - great view of Vancouver, good for photos
go to granville island...take a tour of there ..then take a little ferry 'the aqua bus ' for a 5 minute ride ($2.50) and then walk the seawall around Stanley park...it'll give you an idea of what Vancouver is all about and how beautiful it really is.
Depends what you're after. If you've only got one day and won't be back for a long time, don't go too far. In one day you could manage to see Stanley Park, Gastown, Chinatown, Robson Street, Granville Island, and Kitsalano. It would be a busy day but you'd really get a good overview.

If it's more like a "I need something to do for one day because I've got other things planned for other days" thing, then make a daytrip to Victoria. Get up early and get down to the ferry in Tsawassen. You can even take public transit busses on both sides. Get down to Victoria and see the Parliament Buildings, the Empress Hotel, and the Royal B.C. Museum. At the end of the day head back. Another fulfilling day.

It also depends where you're from. If you're not from the West Coast, try to get in some West Coast cuisine or fast-food: wraps, smoothies, California rolls, B.C. rolls, etc. If you like drinking, try the local beer.

One of my favourite places in Vancouver is the Museum of Anthropology on the U.B.C. grounds. If you're into that thing. Lots of artifacts of native culture. It's right near a ridge that you can walk down to Wreck Beach too.
go to vancouver aquarium and make sure to visit capillano suspension bridge.
Check out the Big Bus Tours... worth every penny and they will take you to all the best places in Van and tell you all about them. You can also get on and off as often as you like - busses show up every 15 minutes at the stops. The pass is good for two days. ...I just did this two weeks ago and thought it was the best thing ever!
On a day trip? What kind of thing interestss you? There are all sorts of activities around the False Creek area, ranging from Science World to the Maritime Museum, with transportation between them provided by little ferry boats. And Granville Island (half way between) is a mecca for arts and crafts and interesting browing through funky little stores.
If you're into outdoor activities you can take the chairlift up Grouse Mountain and hike, or ride mountain bikes, or just have a great dinner and enjoy the view.
For cultural fun go to the Richmond Chinese mall -- just like a visit to Shangahi, and there are Buddhist temples to visit as well.
Check out MyBC.com for more info, and enjoy your day.
