Gastown - historical area of downtown Vancouver, basically Vancouver's first downtown core. Gastown is a mix of tourist-oriented business (generally restricted to Water Street), restaurants, nightclubs, poverty and newly-upscale housing. In addition, there are law firms, architects and other professional offices, as well as computer and internet businesses, art galleries, music and art studios, and acting and film schools.
Gastown's most famous (though nowhere near oldest) landmark is its steam-powered clock, located on the corner of Cambie and Water Street. Chinatown is nearby as well.

Granville Island - major tourist destination, providing amenities such as a public market, a large marina, the Emily Carr Institute of Art and Design (named in honour of the artist), various theatres including one run by The Arts Club Theatre, and various shopping areas.

Stanley Park - a 404.9 hectare, urban park bordering downtown Vancouver. Stanley Park contains numerous natural and man-made attractions, including hiking in the forest, the Vancouver Aquarium, the petting zoo, the miniature train, and the Sea Wall.

Also, if you have time, Whistler is a couple hours a way by car. Whistler is a mountain resort town that's awesome to visit in both summer and winter. There's tons of stuff to do there, especially if you like "outdoorsy" stuff.

Victoria is beautiful too if you have time to take the ferry over to Vancouver Island.
